Involucrin has been the first protein identified as a cornified envelope constituent. Human Involucrin is a 68 kDa protein. In normal human skin, Involucrin expression initiates in the early spinous layer and is maintained in the granular layer.In the transition zone which separates the dead from living epidermal layers, involucrin is incorporated, via the action of transglutaminase, as a component of the cornified envelope. Cross-linked involucrin is deposited around the membrane surface in the vicinity of desmosomes . Involucrin cross-linked to desmosomal proteins functions as a scaffold to which other cornified envelope components are added later by further cross-linking to complete the cornified envelope assembly.
